Phage display was used to select the first Affibody® molecules, binding insulin, Taq DNA polymerase and Apolipoprotein A1 Milano, respectively. Two libraries of potential binders were created by randomization of selected amino acid residues on two helices of of a 58-amino acid domain from Staphylococcal protein A.

Affibody® molecules with extremely high affinity for the tumor antigen HER2 were obtained by affinity maturation based on a previous set of HER2-binders. The novel Affibody® molecules displayed rapid organ distribution kinetics and specific binding to HER2-expressing tumors in vivo in mice, resulting in high contrast gamma camera tumor images within a few hours.

A HER2-binding Affibody® molecule was produced in a single chemical process by peptide synthesis and site-specifically and homogeneously conjugated with a DOTA chelator. Efficient and rapid radiolabeling was achieved and the molecule showed a high, specific tumor uptake as well as a favorable tumor-to-blood ratio in a xenograft model. High-contrast gamma camera images were obtained already 1 hour after injection. Pre-treatment with Herceptin did not interfere with tumor targeting.

In order to achieve a biodistribution pattern suitable for systemic radiotherapy, a HER2-binding Affibody® molecule was fused with an albumin-binding domain (ABD) conjugated with DTPA and labeled with the low-energy beta-emitter (177)Lu. Treatment of HER2-expressing human microxenografts with this conjugate completely prevented formation of tumors, in contrast to treatment with a radiolabeled non-HER2-binding Affibody® molecule. These results demonstrate a potential for treatment of disseminated tumors using Affibody® mediated radiotherapy.

Development of a novel serum albumin binding protein showing an extremely high affinity for HSA, KD in the femtomolar range, corresponding to several orders of magnitude improvement compared with the wild-type domain.
